Ribo Reports Positive Phase 2a Results for Vortosiran in CAD Patients

A 92% reduction in Factor XI activity has been achieved in patients with chronic coronary artery disease using vortosiran, a novel siRNA therapy developed by Suzhou Ribo Life Science. The European study marks the first clinical proof-of-concept for siRNA-mediated FXI suppression, offering a potential long-acting alternative to daily anticoagulants.

The randomized, placebo-controlled trial tested vortosiran in patients already undergoing standard aspirin treatment. Results showed that a 400mg maintenance dose provided sustained FXI suppression for several months, allowing for a dosing interval of three to six months. Researchers noted that this inhibitory effect significantly exceeds that of current small-molecule programs requiring daily administration.

Safety data from the trial showed no treatment-related serious adverse events or major bleeding, addressing a critical hurdle in cardiovascular medicine: preventing clots without compromising hemostasis. Dr. Li-Ming Gan, co-CEO and Global President of R&D, noted that the findings confirm the therapy's potential as a highly differentiated approach to thromboembolic diseases. Following these results, the company has launched the ORBIT-XI program to accelerate the transition into Phase 2b and Phase 3 clinical development.
